The invention discloses an accurate positioning method of an anchor bolt hole of a marine main engine. The method comprises the steps of mounting a channel steel target C at a distance from the rear end of a first anchor bolt hole in the engine before centering a marine axle rudder system; determining the position of an actual center line of an axle system during centering and accepting the axel rudder system; measuring the axial distance from the channel steel target C to the rear end surface of a propeller axle pipe; checking the verticality of the channel steel target C relative to an engine base panel; positioning the axial position of the first anchor bolt hole of the engine through the channel steel target C; checking the axial position of the first anchor bolt hole in the engine, and correspondingly correcting according to the condition. With the adoption of the method, the problem that super poor engine positioning influences the use performance of axle system related accessories, in particular the sealing effect of a rear seal of the propeller axle pipe can be avoided; in addition, the operation is easy, simple and convenient, and does not cause extra initial investment and use cost; the method is suitable for ships of various types, so that the generality is relatively high, and the popularization and application are easy.
