Multidrug resistance (MDR) - an extensive barrier for successful chemotherapies. Currently, P-gp inhibitors in chemotherapeutic treatments attained better results. This paper focuses on critical review which summarizes the clinical and computational significance of P-glycoprotein role in efficacy of drugs in tumor cells, breast cancer, and ovarian cancer along with challenges in the development of P-gp inhibitors towards anti-cancer therapy. This paper covers systematic review of various computational tools in predicting P-gp in human cells in the context of various cancers.
