The vibrant equilibrium of our body, especially the skeletal system, is intricately shaped by the dynamic interplay of the gut microbiota. Antibiotics can directly or indirectly impact the skeletal system by influencing the microbiota. In situations of estrogen deficiency, probiotic bacteria have been documented to impact bone health positively. The relationship between antibiotics, the gut microbiota, and bone health is a complex and developing field of research. This study aims to regulate the in vitro release of azithromycin (AZM) and address challenges linked to oral administration of AZM for influencing gut microbiota and enhancing bone healing. In pursuit of this objective, we formulated AZM-loaded polylactic acid/hydroxyapatite (PLA/HAp) composite microspheres using the double emulsion solvent evaporation technique. The impact of varying HAp contents on the morphology and release profile of PLA/HAp microspheres was also evaluated. The morphology and size of the AZM-loaded microspheres were examined using a high-resolution scanning electron microscope (SEM). The prepared microspheres had a spherical morphology and average particle size of 58.585 µm to 120 µm. Mathematical modelling was conducted to predict the release mechanism of AZM from the composite microspheres. The in vitro AZM release confirmed the reduction of initial burst release from the composite microspheres compared to PLA microspheres and commercially available AZM tablets.
