The present study evaluated the effect of artificial gravity loading on transporter-mediated uptake and release of L-glutamate using the inhibitors of glutamate transporters as tools. Effects of the inhibitors on glutamate transporters are complicated and depend on a wide variety of factors, namely whether conditions favor glutamate uptake or uptake reversal and whether the inhibitor itself is a substrate for the transporters, etc. In summary, the data of the glutamate uptake and carrier-mediated glutamate release suggest that inhibition caused by DL-TBOA and DL-THA increased after centrifuge-induced hypergravity. In this study the authors have attempted to clarify the glutamate transporter modulation process under centrifuge-induced hypergravity, but the whole mechanism still remains largely unexplained. Such knowledge is expected to be of value in designing new therapeutic strategies under different pathological and altered gravity conditions also.
