Surface plasmon resonance is a sensitive method for detecting biomolecular interactions like DNA-DNA, protein-DNA and protein-protein interactions. The principle of this system is to transform a biochemical signal into an optical signal. This method avoids the need of fluorescent labels because we are measuring precisely the local variation of the optical index caused by the biomolecular interaction. This system has been used for many applications including DNA-DNA, protein-DNA and protein-protein interactions and is efficient for every type of interaction if one of the two reagents can be bound to the gold film. After the optimisation of the interacting conditions, it enlightened the specificity of the interaction of the protein with a double strand target DNA. Results will be presented on the affinity of this protein with its target DNA (which are numerous) and on the relative functionality of mutated p53 proteins.
