The invention relates to a device and method for measuring the airtightness of an inclined airtight floor in an aircraft floor structure, and the device comprises an airtight box, an upper cover plate, and a lower cover plate. The inner side of the airtight box is provided with an airtight measurement mounting edge, the upper and lower ends are opened, and the outer walls of the upper and lower ends are respectively provided with an upper concentric-square-shaped skirt edge and a lower concentric-square-shaped skirt edge; the air tightness measurement mounting edge is used for mounting an inclined airtight floor; a part above the air tightness measurement mounting edge in the air tightness box is an air tightness measurement space, and an air pressure loading space is arranged below the air tightness measurement mounting edge; an air pressure detection port, a flowmeter mounting port, a pressurizing air inlet and a pressurizing air outlet are formed in the side wall of the airtight box; the air pressure detection port and the flowmeter mounting port are communicated with the air tightness measurement space; the pressurizing air inlet and the pressurizing air outlet are communicated with the air pressure loading space; the upper cover plate is connected to the upper rectangular-ambulatory-plane skirt edge; and the lower cover plate is connected to the lower rectangular-ambulatory-plane skirt edge.
