The invention discloses a medium-voltage power supply system, a fault positioning method of the medium-voltage power supply system and a motor train unit, one or more coupling contactors of the medium-voltage power supply system are connected to a medium-voltage bus, and each medium-voltage bus section comprises at least one load and at least one auxiliary converter module; each auxiliary converter module comprises an auxiliary converter and a grounding detection module, and the grounding detection module determines whether a grounding fault exists or not. The controller can check whether the auxiliary converter, each section of medium-voltage bus and each load have grounding faults in sequence according to a fixed sequence by controlling the output contactor and the starting contactor, and performs grounding fault detection on each section of medium-voltage bus and the coupling contactor and the load on each section of medium-voltage bus in parallel. Manual troubleshooting one by one is not needed, troubleshooting time and labor cost are greatly saved, and meanwhile the possibility that the load starting contactor and the load are damaged due to improper manual operation is reduced.
