A connection structure of a main shaft and a winding drum of a mine hoist comprises the main shaft, the winding drum, a main shaft flange and a winding drum flange, and the main shaft flange and the winding drum flange have the same thickness and are oppositely arranged inside and outside, so that the main shaft flange and the winding drum flange form an annular flange butt joint assembly used for matched connection. The left side and the right side of the flange butt-joint assembly are each provided with a clamping plate, at least four super-close-fit bolts and a plurality of high-strength bolts are arranged on the annular flange butt-joint assembly in the circumferential direction of the flange butt-joint assembly, and the super-close-fit bolts and the high-strength bolts are perpendicular to each other and penetrate through the flange butt-joint assembly and the two clamping plates. According to the connecting structure of the main shaft and the winding drum of the mine hoist and the connecting method of the connecting structure, the matching geometric dimensions of parts in the connecting structure can be greatly reduced, and the connecting structure is convenient to mount and dismount, large in transmission torque density, good in safety and reliability and particularly suitable for large and medium-sized mine hoists.
