The invention discloses a stratified sampling device for water sample detection and a sampling method. The stratified sampling device for water sample detection comprises a circular floating body, a driving mechanism and a sampling device, the sampling device comprises a telescopic mechanism which is in waterproof sealing connection with the circular floating body, a plurality of negative pressure boxes which are positioned and sleeved on the telescopic mechanism, and a submerged part which is connected with the telescopic mechanism and can extend to a water area outside the circular floating body; and a plurality of water suction ports are circumferentially formed in the telescopic end of the telescopic mechanism. According to the description, the circular floating body rotates and moves to a specified position along the water area through the driving mechanism, the telescopic mechanism drives the submerged part to submerge to the water area with the specified depth, a water sample is sampled in a negative pressure mode, operation is convenient and fast, and the whole process is manually operated on the shore. Besides, the invention provides a stratified sampling method for water sample detection, and the stratified sampling method can cope with different water area environments and can be used for accurately sampling polluted water areas.
