To improve the driving safety and comfort on interchange loop ramps, this paper takes the ramp of Wuxi hub from Nanjing to Jiangyin as an example. After analyzing its security problems, schemes to optimize the horizontal alignment are put forward, including S-curve and oval curve schemes. Through the field investigation and driving simulation, indexes of operating speed and brain waves are applied to evaluate the optimized alignment and the original alignment. The comparison results show that the deceleration effect of an S-curve scheme is more remarkable, and drivers’ tenseness is moderate, which is beneficial to drivers’ vigilance and driving safety. Therefore, the S-curve scheme is better, and also provides a useful reference for the alignment optimization of a long, straight line connected with short-radius turn.
