The goal of this doctoral project was the development of bioactive glasses that can provide antibacterial activity without the addition of any antibiotic drugs in order to be effective against antibiotic-resistance bacteria. In this perspective, non-ordered and ordered silicate mesoporous bioactive glasses (MBGs) were synthesised via conventional sol-gel and surfactant-assisted sol-gel methods, respectively. Antibacterial therapeutic elements (Ag, Cu, Zn, and Ce) in 1, 3 and 5 wt% amounts were incorporated into the MBGs with the base composition of 76SiO2-13CaO-11P2O5 (wt%) in order to provide antibacterial activity. The MBGs obtained by the two different sol-gel methods were also examined for their mesoporous structures, dissolution behaviour, bioactivity and antibacterial properties. Ordered MBGs showed larger surface area, larger pore size and higher pore volume compared to non-ordered MBGs. Among non-ordered and ordered MBGs, Ag-MBGs exhibited the highest dopant release into PBS, which was measured by ICP-OES. The MBG that showed the highest evidence of bioactivity compared to other MBGs was ordered undoped MBG after 72 h immersion in SBF. The most promising MBGs in terms of antibacterial behaviour against E. coli and S. aureus bacterial strains were the non-ordered and ordered Ag-MBGs. Some of the non-ordered MBGs (undoped and 1, 3, and 5 wt% Ag-, Cu- and Zn-MBGs) were also examined for their indirect cytocompatibility behaviour, which was tested with murine fibroblast cell line (L929). The tested extracts were all cytocompatible with L929 cells. Among the other doped MBGs, due to its overall good antibacterial and cytocompatibility behaviour, non-ordered 5 wt% Ag-MBG (NO5 Ag-MBG) was chosen to be combined with a synthetic polymer (poly(ether urethane)) and a natural polymer (chitosan) to make antibacterial polymer/MBG films with potential to be used in wound healing applications.
