A novel biosensor based on surface plasmon resonance (SPR) immunoassay has been developed for the determination of 2,4,6-trinitrotoluene (TNT) and its derivative nitroaromatic explosive compounds. A conjugate immobilized SPR gold sensing surface was used as a solid phase immunoassay and the quantification of TNT was carried out based on the principle of indirect competitive immunoreaction. Three types of anti-2,4,6-trinitrophenol (TNP) antibodies (polyclonal antibody prepared in the authors' laboratory, commercial polyclonal and monoclonal antibodies) were used for immunoreaction with TNP-bovine serum albumin conjugate (TNP-BSA) and the results were compared. The results showed that detection sensitivity down to 60 ppt of TNT could be achieved with the proposed immunoassay system. The cross reactions to related nitroaromatic signature compounds, which can exist with TNT at the landmine sites have been evaluated. The results clearly indicate the potential of the proposed immunoassay for the development of simple and user-friendly sensor system for on-site detection of landmines.
