An electro-hydraulic brake assembly (20) comprises a hydraulic control unit (HCU) body (30) having a top surface (34) and a bottom surface (36) opposite the top surface (34) defining a master cylinder bore (57) and a pressure supply bore (125). A fluid reservoir (50) is disposed on the top surface (34). A primary piston (66) is slidably disposed in the master cylinder bore (57) and configured to supply brake fluid to a wheel brake (22, 24, 26, 28) in response to pressing of a brake pedal (80). A pressure supply unit (128) includes: a pressure supply piston (190) disposed within the pressure supply bore (125); a motor (200) located on the bottom surface (36) of the HCU body (30) and having a motor shaft (212). An actuator mechanism (194) includes a threaded shaft (196) configured to be rotated by the motor shaft (212), and a nut (198) coupled to the pressure supply piston (190), together causing the pressure supply piston (190) to translate linearly through the pressure supply bore (125) in response to rotation of the motor shaft (212).
