The invention discloses an intelligent locker automatic replacement system and method of a transportation robot. The system comprises an intelligent locker, the transportation robot and a locker storage area. The intelligent storage cabinet is provided with a first control module and one or more storage chambers with cabin doors, and each storage chamber is provided with a locking device used for locking the corresponding cabin door. The transportation robot comprises a vehicle body, a second control module arranged on the vehicle body, a controller, a reader, a lifting mechanism and a storage table. The storage cabinet storage area is provided with a plurality of storage platforms, the storage platforms are provided with placing tables and parking areas, and the transportation robot can place and carry the intelligent storage cabinets to the placing tables or move the intelligent storage cabinets away to the placing tables so that the intelligent storage cabinets in different layout forms can be replaced. According to the intelligent locker automatic replacement system of the transportation robot, the transportation robot can selectively unload the carried intelligent locker to the no-load storage platform and replace the intelligent locker with the corresponding layout specification according to requirements, so that the logistics distribution efficiency is improved.
