Along with the quick development of highway passenger transportation in comprehensive transportation, the slow-evolving management theory and methods of highway passenger transportation could not meet the demands of the change, which leads to the low efficiency and poor service quality, revealing the irrationality and no scientificalness. Under the consideration of Whole Trip Course and the Relative Inefficient Fare, by the calculation of the time and fare of the highway trip in microcosmic, the paper aims at establishing the multi-objective optimization model, putting forward some specific measures and example practice to follow. The result has been used on the transport line regulation in a main hub effectively by the calculations of the actual lines, which exerts a nonreplaceable role for the rapid and smoothly transport service as a part of the public transportation management.
