Biological analysis based on PCR and clone library approache, along with physical analytical techniques (XRD, XRF, γ-Spectrometry) were used to characterise 'black powder', a major constituent of the environmentally hazardous wastes generated from the natural gas industry. Black powder is a very complex material, containing hydrocarbons, Sulfates and iron oxides. In addition, microbial communities can often be associated with these materials. A sample was collected from a natural gas pipeline, and 16S rRNA sequenced to characterise the indigenous microbial community. The microbial community (29 clones) was dominated by bacterial species identified as Iron-reducing bacterium enrichment culture clone HN54 (24.14% of 16S rRNA clone library), Clostridium sp. enrichment culture clone tpsrb007 (17.24% of 16S rRNA clone library) and Uncultured bacterium clone 227 16S (34.48% of 16S rRNA clone library). Magnetite (Fe3O4) was the most abundant mineral phase, while γ-spectroscopy did not show any unusual radioactivity content.
